HPLC Chiral Separation of Praziquantel on Cyralis CM Column
June 17, 2026
HPLC Method for Analysis of Praziquantel on Cyralis CM Column by SIELC Technologies
High Performance Liquid Chromatography (HPLC) Method for Analysis of Praziquantel.
Praziquantel is an drug with the chemical formula C19H24N2O2It is primarily used to treat parasitic worm infections in a variety of living beings. In humans, it is used to treat schistosomiasis. tapeworm infections, cysticercosis, and more. It works through increasing the permeability of the membranes of schistosome cells towards calcium ions which leads to contractions of muscles and therefore paralysis of the parasite. You can find detailed UV spectra of Praziquantel and information about its various lambda maxima by visiting the following link.
This application describes a normal-phase chiral HPLC method for the separation of Praziquantel enantiomers using SIELC’s Cyralis CM chiral column. Cyralis CM is a cellulose-based chiral stationary phase (CSP) designed for enantiomeric separations, optical purity testing, chiral impurity analysis, and stereoisomer characterization in pharmaceutical, chemical, agrochemical, and fine chemical applications.
Praziquantel can be retained and analyzed using the Cyralis CM stationary phase column. The analysis utilizes an isocratic method with a mobile phase consisting of Hexane and Ethanol. Detection is performed using UV.

For optimal results in HPLC analysis, it is recommended to measure absorbance at a wavelength that matches the absorption maximum of the compound(s) being analyzed. The UV spectrum shown can assist in selecting an appropriate wavelength for your analysis. Please note that certain mobile phases and buffers may block wavelengths below 230 nm, rendering absorbance measurement at these wavelengths ineffective. If detection below 230 nm is required, it is recommended to use acetonitrile and water as low UV-transparent mobile phases, with phosphoric acid and its salts, sulfuric acid, and TFA as buffers.
For some compounds, the UV-Vis Spectrum is affected by the pH of the mobile phase. The spectra presented here are measured with an acidic mobile phase that has a pH of 3 or lower.

Legacy L1
SIELC's family of Legacy columns is based on the United States Pharmacopeia's (USP) published chromatographic methods and procedures. Numerous brands have columns used in USP reference standards and methods. USP has created various designations to group together columns with similar types of packing and properties in the solid phase. SIELC's Legacy columns adhere to these strict requirements and properties, allowing you to easily replace older columns that are no longer available without needing to significantly modify your method or SOPs.
