Primesep 200 separates the very hydrophilic cotinine metabolites. Ion-exchange and other polar interactions provide retention for a compound that is difficult to retain on traditional C18 reversed-phase columns. The separation uses a simple mobile phase of water, acetonitrile (MeCN, ACN), and trifluoracetic acid (TFA) with UV detection.
