HPLC Method for Analysis of Streptomycin on Chromni Column by SIELC Technologies

High Performance Liquid Chromatography (HPLC) Method for Analysis of Streptomycin.

Streptomycin is the an antibiotic with the chemical formula C21H39N7O12. It is primarily used to treat bacterial infections but not limited to tuberculosis, endocarditis, brucellosis, and the plague. It is typically given intramuscularly. It works as a protein synthesis inhibitor by binding to the 16S rRNA of the 30S ribosomal subunit, which leads to it interfering with the binding of formyl-methionyl-tRNA. It is the official state microbe of New Jersey. You can find detailed UV spectra of Streptomycin and information about its various lambda maxima by visiting the following link.

Streptomycin can be retained and analyzed using the Chromni stationary phase column. The analysis utilizes a gradient method with a simple mobile phase consisting of water and acetonitrile (MeCN). Detection is performed using LC MS.

For optimal results in HPLC analysis, it is recommended to measure absorbance at a wavelength that matches the absorption maximum of the compound(s) being analyzed. The UV spectrum shown can assist in selecting an appropriate wavelength for your analysis. Please note that certain mobile phases and buffers may block wavelengths below 230 nm, rendering absorbance measurement at these wavelengths ineffective. If detection below 230 nm is required, it is recommended to use acetonitrile and water as low UV-transparent mobile phases, with phosphoric acid and its salts, sulfuric acid, and TFA as buffers.
For some compounds, the UV-Vis Spectrum is affected by the pH of the mobile phase. The spectra presented here are measured with an acidic mobile phase that has a pH of 3 or lower.

HPLC Determination of Streptomycin sulfate salt on Primesep 200 Column

High Performance Liquid Chromatography (HPLC) Method for Analysis of Streptothricins, Streptomycin B, Streptomycin.

Streptomycin is an antibiotic used in treating bacterial infections and has also been used as a pesticide. It is on the World Health Organization’s (WHO) list of essential medicines. It works by inhibiting the synthesis of proteins. It can be retained on a Primesep 200 mixed-mode column by hydrophobic and ion-exchange properties. The analytical method uses MS-compatible mobile phase of acetonitrile (ACN) and water with sulfuric acid (H2SO4) buffer. Also capable of UV detection at 200nm.

Condition

Column Primesep 200
Mobile Phase MeCN/H2O – 10/90%
Buffer H2SO4 – 0.05%
Flow Rate 1.0 ml/min
