High Performance Liquid Chromatography (HPLC) Method for Analysis of Ofloxacin
Ofloxacin is a common antibiotic with the chemical formula C18H20FN3O4. It is typically taken by mouth or injected into a vein to treat variety of bacterial infections. Those infections include pneumonia, prostatitis, and the plague. As eye and ear drops, it is also used to treat superficial bacterial infections of the eye and otitis media. As a fluoroquinolone, it is well tolerated, but may cause nausea, vomiting, and diarrhea as a side effect.
Ofloxacin can be measured at low UV. Using a Primesep D reverse-phase column and a mobile phase consisting of water and an acetonitrile (MeCN) gradient with a sulfuric acid (H2SO4) buffer, Ofloxacin can be retained. This analysis method can be UV detected at 253 nm.

Ofloxacin is a synthetic second generation fluoroquinolone antibacterial agent that was previously used for treatment of bacterial infections. It was later replaced by less toxic medications, because Ofloxacin is harmful if swallowed and when heated to decomposition it emits toxic fumes. Primesep 200, a reverse phase column, contains embedded acidic ionizable groups and can retain Ofloxacin. The method is UV compatible and can be used as a general approach for analyzing similar compounds.
