High Performance Liquid Chromatography (HPLC) Method for Analysis of Trimipramine, Trimipramine maleate.

Trimipramine is a tricyclic antidepressant (TCA) with the chemical formula C20H26N2. It is used primarily used to treat depression, but it has also been used to treat insomnia, anxiety disorders, and psychosis due to it’s sedative, anxiolytic, and antipsychotic effects. Unlike other TCAs, it is a weaker monoamine reuptake inhibitor. This application describes a normal-phase chiral HPLC method for the separation of Trimipramine, Trimipramine maleate enantiomers using SIELC’s Cyralis CM chiral column. Cyralis CM is a cellulose-based chiral stationary phase (CSP) designed for enantiomeric separations, optical purity testing, chiral impurity analysis, and stereoisomer characterization in pharmaceutical, chemical, agrochemical, and fine chemical applications. For best results, the sample should be prepared fresh for analysis.

Trimipramine, Trimipramine maleate can be retained and analyzed using the Cyralis CM stationary phase column. The analysis utilizes an isocratic method with a mobile phase consisting of sodium perchlorate and acetonitrile. Detection is performed using UV.

It acts by decreasing the reuptake of norepinephrine and serotonin (5-HT), which is why it is used as treatment of major depressive disorder.
