PBTC (2-phosphonobutane-1,2,4-tricarboxylic acid) is one of the most widely used scale inhibitors in the cooling water treatment industry.  PBTC can be detected at low UV.  Using Newcrom BH mixed-mode column and a mobile phase consisting of acetonitrile (ACN) and water with phosphoric acid (H3PO4) buffer, 2-phosphonobutane-1,2,4-tricarboxylic acid can be retained and UV detected at 220nm.

Separation of 2-Phosphono-1,2,4-butanetricarboxylic acid on Newcrom C18 HPLC column

2-Phosphono-1,2,4-butanetricarboxylic acid can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
