Piracetam and Levetiracetam are two similar compounds in the racetam family with two varying degrees of effects on the body. Piracetam has been marketed as a treatment for myoclonus and as a way to enhance cognition, although neither claim is approved by the FDA. Levetiracetam, on the other hand, is a common treatment for epileptic seizures (and approved by the FDA).
These racetams can be detected in the low UV regime. There are two effective methods for retaining, separating, and analyzing these compounds. The first uses a SHARC 1 hydrogen-bonding column and a mobile phase consisting of acetonitrile (MeCN) and water without a buffer. The second uses Newcrom R1 mixed-mode column and a mobile phase consisting of acetonirile (MeCN) and water with a sulfuric acid (H2SO4) buffer. Either analysis method can be UV detected at 200 nm with high resolution.

Separation of Levetiracetam on Newcrom R1 HPLC column

Levetiracetam can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
